Disease Based Computational Drug Repurposing: A Review

Drug discovery is a slowly progressing activity in conventional drug development. Apart from this, drug repurposing, an emerging research area in this field, is of less risk, economically feasible, and fast. There are two drug repurposing principles namely, drug-based and disease-based. Here, we are reviewing different disease-based re-purposing using computational methods, which makes use of existing drugs for curing different target diseases. This paper initially gives an outline of the approaches and strategies of drug repurposing and network theory. It then moves to various computational drug re-purposing methods, focussing on network-based and machine learning-based models. Finally, it concludes by discussing various proposed and promising models in this aspect of computational drug re-purposing. We suggest this review as strong foundation that will help the scientists working in computational pharmacology.
